• Description: Recombinant RSV (strain Long) Fusion glycoprotein F0 is produced in a Mammalian expression system. The target gene encoding Gln26-Leu513 is expressed with a C-terminal 6His tag. Stabilizing mutations are introduced to maintain the trimeric prefusion conformation of the RSV Fusion glycoprotein.

• Introduction: Respiratory syncytial virus (RSV) is a highly contagious respiratory pathogen, significantly affecting infants and the elderly. Given the substantial disease burden, an effective vaccine remains a critical unmet need. The RSV fusion glycoprotein (RSV F) is a primary vaccine target as it elicits the majority of neutralizing antibodies. RSV F facilitates viral-host membrane fusion through an irreversible conformational change from a metastable prefusion state to a stable postfusion state. Similar to other Class I fusion proteins, the inactive precursor F0 is cleaved by furin-like proteases into F2, p27, and F1 polypeptides, with F1 containing the hydrophobic fusion peptide. Although both conformations share neutralizing epitopes, those specific to the prefusion F (pre-F) are significantly more potent. Stabilizing the pre-F structure is a key strategy in structure-based vaccine design, offering promise for a safe and effective RSV vaccine.
