| Introduction: | Monkeypox is a zoonotic disease caused by the monkeypox virus (MPXV), an orthopoxvirus. The MPXV E8L protein is homologous to the vaccinia virus D8L protein, which binds cellular chondroitin sulfate (CS), facilitating a distinct virus-GAG interaction. As an abundant 32-kDa IMV protein, D8L is crucial for virus adsorption and entry. D8L elicits strong humoral and cellular immune responses; antibodies against it show neutralizing activity in vitro, while soluble D8L inhibits viral adsorption. DNA vaccines encoding D8L provide partial protection against lethal vaccinia challenge in mice and enhance the efficacy of polyvalent vaccines. |
