| Introduction: | Monkeypox is a zoonotic disease caused by the monkeypox virus (MPXV), an orthopoxvirus. The A29L protein is an intracellular mature virion (IMV) surface envelope protein homologous to the vaccinia virus (VACV) A27 and is conserved across Poxviridae. The A27L protein (p14) is multifunctional, implicated in viral attachment, fusion, and plaque formation. A27 presence in IMVs inhibits fusion protein A26 assembly. Following endocytosis, acidic conditions trigger A26 dissociation, allowing viral-vesicle membrane fusion. A27 anchors A26 to the mature virus (MV) by interacting with the integral membrane protein A17 and forming a disulfide-linked complex with A26. |
