| Introduction: | Human parainfluenza virus type 3 (HPIV-3) is a major cause of lower respiratory tract infections in children. Viral entry and spread are initiated by the hemagglutinin-neuraminidase (HN) and fusion (F) envelope glycoproteins. HN binding to cellular receptors triggers conformational changes in F, leading to virus-cell fusion. The F protein is a type I integral membrane protein with an ectodomain, transmembrane anchor, and cytoplasmic tail. Synthesized as an inactive F0 precursor, it is cleaved into F1 and F2 subunits. The trimeric F structure exists on infectious virions in a metastable prefusion conformation with the hydrophobic fusion peptide buried internally. |
