| Introduction: | The coronavirus spike (S) glycoprotein is essential for viral attachment to host cells at the onset of infection, particularly in severe acute respiratory syndrome (SARS). The SARS-CoV S glycoprotein alone mediates membrane fusion required for viral entry and cell fusion. It acts as a major immunogen and a target for entry inhibitors. Studies indicate that 2019-nCoV infects human respiratory epithelial cells via the ACE2 receptor. The S protein is a large type I transmembrane protein consisting of S1 and S2 subunits. S1 contains the receptor-binding domain (RBD) for receptor recognition, whereas S2 facilitates membrane fusion. The S protein is key to inducing neutralizing antibodies, T-cell responses, and protective immunity. |
