Product Name: | Prostate Specific Membrane Antigen (PSMA) |
Description: | Prostate Specific Membrane Antigen (PSMA) (Lys44-Ala750) is CHO-derived human PSMA/FOLH1/NAALADase I protein and fused with an N-terminal 6-His tag. |
Species: | Human |
Molecular Weight: | Predicted Molecular Mass: 80 kDa; SDS-PAGE: 110 kDa, reducing conditions. |
Source: | CHO |
Introduction: | The enzyme FOLH1 is the target for the antibody-based drug Capromab, which is used for diagnosis of prostate cancer and detection of intra-pelvic metastases. FOLH1 has both folate hydrolase and N-acetylated-alpha-linked-acidic dipeptidase (NAALADase) activity and is involved in prostate tumor progression. Immunohistochemical staining with the antibody HPA010593 shows strong staining of glandular cells in the prostate. PSMA is a cancer antigen that is widely expressed as a noncovalent homodimer on the surface of prostate cancer cells, rather than on normal cells, therefore it is an interesting target to image prostate tissues when being used with radioactive antibodies. PSMA is expressed on the surface of novel blood vessels that nourish a broad range of other solid tumors. PSMA is a type II membrane protein with folate hydrolase activity produced by prostatic epithelium. PSMA expression is found in extraprostatic tissues, including small bowel and brain. |
Purity: | >95%, by SDS-PAGE visualized with Silver Staining and quantitative densitometry by Coomassie® Blue Staining. |
Biological Activity: | Measured by its ability to hydrolyze the substrate N-acetyl-L-Asp-L-Glu into N-acetyl-L-Asp and L-Glu. The L-Glu product is measured by fluorescence after its derivatization by ortho-phthaldialdehyde. The specific activity is >400 pmol/min/µg, as measured under the described conditions. |
Application: | The enzyme FOLH1 is the target for the antibody-based drug Capromab, which is used for diagnosis of prostate cancer and detection of intra-pelvic metastases. |
