After dominating the medicine catalog for decades, small molecules are gradually giving market shares to a second, fast-growing class of drugs – therapeutic proteins. Although the former is currently the main therapeutic agent by number, the impact of protein therapeutics is increasing thanks to advances in recombinant DNA technology and improved understanding of disease pathology. These recombinant therapeutic proteins are also known as biotechnological therapeutics and used for multiple purposes, including diagnosis, prophylaxis, disease management, and/or cure. Moreover, protein therapeutics are known to represent one-third of the entities that are launched as new medicines under biologics.

Nowadays, the importance of therapeutic proteins as an important class of medicines cannot be ignored by patients or the health care industry. Recently approved protein therapeutics attach a wide range of clinical disorders, including cancers, autoimmunity/inflammation, exposure to infectious agents, and genetic disorders, which are difficult to treat by conventional means.

Since 2011, the U.S. Food and Drug Administration has approved 62 recombinant therapeutic proteins, of which, over half of the approved therapeutic proteins were indicated for oncology (26%) and hematology (29%), whereas the remaining 45% had primary indications in cardiology/vascular disease (5%), dermatology (3%), endocrinology (6%), immunology (6%), infectious diseases (3%), musculoskeletal (8%), ophthalmology (3%), pulmonary/respiratory disease (3%), and others.For instance, IL-1Ra is used to treat metabolic disorders or endocrine dysfunctions like type 2 diabetes mellitus and hemophilia, interferon-alpha is used for the treatment of hepatitis C and erythropoietin for chronic anemia, botulinum toxin type A and B are used for multiple dystonia and lepirudin for heparin-induced thrombocytopenia.
